Molecular Charecterization of Melissococcus pluton of hive honeybees

Studied the inlidence of European foul brood disease in relation to temperature and relative humidity in Apis mellifera & A.cerana produced the antiserum against castual bacterium Melissococcus pluton of A.mellifera and detected M.pluton through polymerase chain reaction technique. The molecular cliversity of 8 nucleotides between the isolates was established through ITS (intergenic Transcribed Spacer) primers. Sequenced the 812/550 bps of bacterial isolates and sequences were submitted to the web in European molecular Biology laboratory-European bio informatics Institute (EMBL EBI) Cambridege, UK database and got accession number FN297846 for A.mellifera and FN297845 for A.cerana, M.pluton amplicons. Established phylogenetic relationship of present M.pluton isolates with other reported M.pluton strains with Genbank database. Results revealed that the present isolates were closely related to German stain Melissococca pluton (LTH-3442-AJ). Evaluated 22 antibiotics invitro and ciprofloxacin antibiotic invivo conditions. Detected the ciprofloxacin residues in honey through LSMS chromatography at Indian Institute of Integrative medicines (IIIM) Jammu.
