Details

The aim of this study was to evaluate the morphophysiological behavior of the cultivars Vitis vinifera and Vitis riparia when grown under different stress conditions in vitro. To this end, three methods of leaf pigment extraction were investigated, as well as the effect of different seals and increasing concentrations of sucrose, the effect of ethylene gas and, finally, five levels of irradiance and two culture media, in hyoperidic and normal plants. Extraction of leaf pigments using DMSO is as efficient as using 80% acetone for the extraction of chlorophylls a and more efficient for the extraction of Chl b for V. vinifera grown in vitro and more efficient for the extraction of chlorophylls a and b for both cultivars acclimatized ex vitro. Increasing sucrose concentrations and gas exchange permeable seals had favorable effects on plant growth parameters. Both cultivars were significantly affected when grown in medium containing the ethylene precursor. High levels of irradiance favor alterations and damage to the structures of hyperhydric and normal plants.
