Pulsincap DDS for Chronotherapy
Details
The present work showed that pulsatile release of bambuterol HCL was successfully formulated by pulsincap drug delivery system. In formulating pulsatile drug delivery, the polymers used was hydrophilic polymer showing erodible behavior i.e. low viscosity grade HPMCK4M. for hydrogel plug. The sodium starch glycolate were used, which creates internal pressure on hydration and achieves the bursting effect. On the basis of 32 factorial design 9 batches were formulated, from F3, F6, F9 Contains 200 mg of HPMCK4M. in these formulations, the required lag time was not achieved i.e. 6 hour because of higher concentration of HPMCK4M. In case of F2, F5, F8 the concentration used were 175mg of HPMCK4M. these formulations showed the drug release from 4 hrs up to 9 hrs so these batches also were unable to show significant lag time.Now, in the F1, F4, F7 the concentration HPMCK4M used were 150 mg. in case of these batches the results obtainted were satisfactory and here the required lag time was also achieved, among these three batches f4 was selected as an optimized batch, as it showed the maximum release i.e. up to 61% in 6 hrs and rest of the drug was rest of the drug was released in 8 hr.
