Selenite broth is used as an enrichment medium for the isolation of Salmonella species. This medium must not be autoclaved. Once prepared it is steamed at 100°C for 30 minutes. There should be a very slight red precipitate. To minimise the risk of teratogenicity to workers, sodium selenite must be added separately to the medium. It has a pH of approximately 7.1. Salmonella is a genus of rod-shaped, Gram-negative, non-spore-forming, predominantly motile enterobacteria with diameters around 0.7 to 1.5 µm, lengths from 2 to 5 µm, and flagella which grade in all directions (i.e. peritrichous).
